 jake Sully (Sam Worthington) adalah seorang mantan angkatan laut Amerika Serikat yang kedua kakinya lumpuh dikarenakan cidera tulang belakang. Ia terpilih untuk berpartisipasi dalam program Avatar untuk menggantikan kembarannya yang meninggal dunia. Proyek yang memungkinkannya bisa berjalan kembali. Jake menuju ke Pandora, sebuah bulan nan subur yang penuh dengan berbagai macam makhluk hidup. Pandora merupakan habitat bagi suku Na’vi, makhluk yang mirip manusia dengan kehidupan primitif serta memiliki kemampuan seperti manusia. Saat manusia mencoba memasuki Pandora untuk menambang mineral bernama unobtanium yang ada di sana, suku Na’vi memerintahkan para prajuritnya untuk melindungi negerinya dari ancaman. Jake direkrut untuk menjadi bagian dari proyek ini. Dikarenakan manusia tidak dapat menghirup udara di negeri Pandora, maka mereka menciptakan makhluk yang mirip suku Na’vi yang mereka sebut sebagai Avatar. Dengan tubuh Avatar, Jake dapat berjalan kembali. Di hutan Pandora, Jake melihat banyak keindahan dan bahaya. Ia juga bertemu dengan wanita muda dari suku Na’vi bernama Neytiri (Zoe Saldana). Seiring berjalannya waktu, Jake berbaur dengan suku Na’vi dan jatuh cinta kepada Neytiri. Pada akhirnya, Jake dilanda dilema antara tujuannya dikirim oleh perusahaan tambang ke Pandora, dan suku Na’vi yang memaksanya untuk memihak pada satu pilihan yang akan menentukan nasib suku Na’vi dan manusia di Pandora
